Effect of Ethionine on RNA and Protein Syntheses of Regenerating Liver in Rats

Abstract
The present paper was to observe the effect of acute ethionine intoxication on the RNA and protein synthesis of regenerating rat live in vivo and in vitro.
The study was also extended to the regenerating livers of chronically ethionine introxicated rats.
In vivo experiment, intraperitoneal administration of ethionine caused a striking inhibition of orotic acid-^(14)C incorporation into liver RNA at an equal rate in non-hepatectomized and partially hepatectomized rats, whereas the inhibition of L-lysine-^(14)C incorporation into the liver and serum protein was considerably less significant in the regenerating liver.
In the liver slice experiment, the rises in RNA and protein syntheses after removal of part of liver were more significant compared to the rises observed in vivo.
The inhibitory effect of ethionine, in vitro, was more remarkable on the synthesis of RNA than that of protein, but there was no difference in the degree of inhibitions between non-hepatectomized and regenerating livers.
Protein synthesis of ethionine induced cirrhotic livers showed a considerable increase after partial hepatectomy in vivo.
The regenerating liver of the chronically ethionine intoxicated rats was more strongly inhibited in protein synthesis by a single large dose of ethionine than regenerating liver of normal rats.
